TramaLa serie è ambientata a New York, nella sede dell'ISIS (International Secret Intelligence Service), agenzia di spionaggio fittizia dove lavora il protagonista Sterling Malory Archer, spia dal nome in codice Duchessa. Sterling Archer è molto abile nel suo lavoro, ma anche estremamente egocentrico ed egoista, dedito ad un edonismo sfrenato, alcolista incallito e burlador provetto.
Gli agenti sul campo dell'ISIS sono Lana Kane e Ray Gillette, gli altri personaggi sono Cyril Figgis, contabile, Pam Poovey, direttrice delle risorse umane, il dottor Algernop Krieger, capo della sezione Ricerca e Sviluppo dell'ISIS, e Cheryl Tunt, segretaria. Ad essi si aggiunge il capo dell'ISIS, la madre di Sterling, Malory Archer, boss molto dominante, alcolista come il figlio.
